BioDuro’s biology team is headed by an experienced group of biologists who provide a broad array of quality services as well as in-depth biological support for integrated drug discovery programs. Our modern laboratories are fully equipped to quickly deliver a variety of services.
Reagent Generation
- Gene to protein: cloning, expression (E. coli, baculovirus and mammalian) and protein purification (for assay and crystallography)
- Cell-line construction (clone, express and characterize gene of interest)
- Antibody reagents or therapeutics
Assay Development
- Biochemical assays (enzyme, receptors)
- Cell-based assays
- Human whole blood assay and human primary cell-based assays
- Biomarker and ex-vivo assays
- Assay detection formats include UV-Vis, fluorescence, FP, HTRF, filter binding, SPA and other radiometric assays, luminescence, AlphaScreen, LC/MS, FLIPR, ELISA and FACS
Screening
- HTS and hit characterization including MOA and kinetic determination
- Primary, secondary and selectivity screening for lead optimization
Mechanistic Biochemistry and Toxicology
- Mechanism of action determination for enzyme inhibitors
- Mechanistic pharmacology for GPCR agonists and antagonists
- On- and off -rate determinations
- Characterization of compound binding sites
- Biotransformation and bioactivation/inactivation of compounds: e.g. CYP, GST, UGT
- Detection of covalent binding of reactive metabolites
In addition, our scientists have extensive experience in using siRNA to characterize cellular function of target genes. Combined with our disease model pharmacology expertise in oncology, metabolic disease and immunology, we can carry out target validation studies that are tailored to the needs of our clients.
